I’m working with a big plasmid \~17kb. The first time I miniprepped it with the genejet miniprep kit, I got a great yield turned out to be mostly gDNA contamination, which I’m assuming is why my restriction digest failed, but there was a band about the size of the plasmid.

I tried miniprepping again and got a more reasonable yield, redid the restriction digest and it still failed and there is streaking present.

The plasmid contains a big gene cluster that seems to be slowing down the growth of the culture so maybe that related?

Idk I’d love anybody’s advice!
