I am trying out a basic IPS Samsung monitor here. It has a saturation slider in the picture settings in addition to the individual R, G, B sliders.

The R, G, B sliders are for channel gain and default to 50. The saturation slider also defaults to 50, but I am finding that it looks a bit more "natural" at 40-44. It's not a convert to greyscale slider, just desaturates.

Wondering how calibration would take all this into account? I know R, G, B would set the white point.

Is it that you just calibrate with it at the default of 50, and then adjust the saturation to taste after?
